From separation to optical detection in radially elongated pillar columns


Chromatography is the most prominent analytical separation technique in life sciences, diagnostics, pharmacy, etc. An important limitation on the attainable separation performance is set by the disordered nature of the packing. In this work micro-machined and thus perfectly ordered radially elongated pillar array (REP) columns were studied. It has been demonstrated that this novel column format can outperform state-of-the-art columns.

In miniaturized chromatography columns there are today already some optical detection techniques like UV-VIS detection commercially available in flow cell volumes below 20nl. For the characterization of macromolecules multi-angle laser light scattering (MALLS) is highly relevant however it cannot be implemented today as it is not compatible with pillar arrays. In this PhD a scatter cell with a volume as low as 15nl has been investigated for MALLS detection.
